A woman has died and another person is in hospital with serious injuries after a fire broke out at a backpacker hostel in East Perth overnight.

A man is assisting police with their investigation into the blaze, which is being treated as suspicious.

Emergency services received a triple-zero call just after midnight, reporting flames from the two-storey Exclusive Backpackers hostel on Adelaide Terrace.

Residents were evacuated shortly after as firefighters worked to contain the fire to the top floor.

More than a dozen people were left waiting on the street, many with little more than the shirts on their backs.

A line of people sit on a footpath next to some ambulances and fire trucks.

One woman found seriously injured inside the hostel was taken to hospital, where she later died.

Ambulance crews took two people to Royal Perth Hospital, where one person remains in a serious condition.

A further three people were treated at the scene for smoke inhalation.

The cause of the fire is not clear, but police arson and homicide squads are investigating, with detectives are speaking to other people who were staying at the hostel.
